What is Retatrutide?

Retatrutide (LY3437943) is a synthetic 39-amino-acid peptide developed by Eli Lilly and Company that acts as an agonist at three receptors: the glucose-dependent insulinotropic polypeptide (GIP) receptor, the glucagon-like peptide-1 (GLP-1) receptor and the glucagon receptor [1]. Its sequence contains the non-coded amino acids α-aminoisobutyric acid (Aib) and α-methyl-L-leucine and ends in a C-terminal amide. Lysine 17 carries a C20 fatty diacid attached through a γ-glutamic acid–AEEA linker, a modification that confers albumin binding.

Its receptor pharmacology has been characterized in cell-based signaling assays, where it shows balanced glucagon and GLP-1 receptor activity and greater GIP receptor activity, and it has been studied in mouse models of energy balance and glucose metabolism [1].
